A Hierarchical Network Controls Protein Translation during Murine Embryonic Stem Cell Self-Renewal
and Differentiation

Stem cell differentiation involves changes in transcription, but little is known about translational control during differentiation. The authors comprehensively profiled gene expression during murine embryonic stem cell (ESC) differentiation into embryoid bodies by integrating transcriptome analysis with global assessment of ribosome loading. They show that a hierarchy of translational regulators, including mTOR, 4EBP1, and the RNA binding proteins DAZL and GRSF1, control global and selective protein synthesis during ESC differentiation. The combination of limited protein synthesis while in the pluripotent state with hierarchical translational regulation during differentiation may provide a quality control mechanism during self-renewal and fate choices of ESCs.

Three-Year Post-Doctoral Fellowship

We seek a postdoctoral fellow with experience in working with human ES cells to join a privately-funded laboratory in Manhattan associated with Columbia University (investigators: Drs. H. Wichterle, T.M. Jessell and C.E. Henderson). The candidate will use motor neurons derived from new ES cell lines to study disease mechanisms in ALS and SMA. The work will be carried out in collaboration with scientists at the Harvard Stem Cell Institute and the Memorial Sloan-Kettering Cancer Center. The fellowship is for 3 years.
